Biofuncionalização de titânio com plantago major: citocompatibilidade, mineralização e modificação de superfície in vitro

Resumo

Este estudo avaliou o uso do extrato alcoólico de Plantago major como agente de biofuncionalização de superfícies de titânio, com foco na caracterização físico‑química, citocompatibilidade e mineralização in vitro. O extrato foi caracterizado por DLS e potencial zeta, apresentando sistema coloidal heterogêneo, com polidispersidade elevada e carga superficial negativa, além de grupos funcionais polares identificados por FTIR; a morfologia particulada e a formação de aglomerados foram observadas por microscopia eletrônica de varredura. A citocompatibilidade foi avaliada por MTT em células-tronco mesenquimais, MC3T3‑E1 e SaOS‑2, mostrando manutenção de orientações compatíveis com a ISO 10993‑5 em concentrações até 0,1 mg/mL, com respostas dependentes de dose, tempo e linhagem celular. A mineralização da matriz extracelular, avaliada por Alizarina Red S após 14 dias, foi aumentada de forma pronunciada, especialmente em 0,05 mg/mL. A biofuncionalização do titânio foi confirmada por FTIR e MEV, com aumento expressivo de hidrofilicidade (redução do ângulo de contato) e presença de material orgânico aderido à superfície. Na interface titânio–extrato–célula, o ensaio CCK‑8 indicou ausência de citotoxicidade associada à funcionalização, embora não tenha sido adicionado ganho de rotina em relação ao titânio não funcionalizado. Em conjunto, os achados apoiam o potencial do extrato como modificador superficial de titânio, com efeitos pró‑mineralizantes em modelos osteoblásticos in vitro.
